Berberine (berberine) is mainly extracted from plants such as Coptis chinensis, Phellodendron amurense, and Huangbo tree roots. Here is a comparison of several typical methods:
1. Microwave assisted extraction method
·Core principle: Using microwave heating to penetrate cells, destroy structures, and accelerate the dissolution of active ingredients.
·Key steps/parameters: After soaking Huanglian powder in water, microwave extract for 3 minutes at a power of 500W, with a solid-liquid ratio (weight to volume ratio of raw materials to water) of 1:120, and extract twice.
·Advantages and outputs: fast extraction speed, high efficiency, and environmental friendliness. Research shows that the extraction rate can reach 5.61%.
2. Acid water/acid-base salt precipitation method
·Core principle: Use acid solution to extract alkaloids, and then precipitate them by adding salt (such as sodium chloride) or adjusting the pH.
·Key steps/parameters: Soak the raw materials in sulfuric acid or hydrochloric acid, adjust the pH to 1-2, add salt precipitation, or further purify using zinc chloride, lime water, etc.
·Advantages and outputs: The process is relatively mature. The content of berberine in a finished product can reach 98.1%.
3. Organic solvent (ethanol) reflux extraction method
·Core principle: Using organic solvents such as ethanol to heat and reflux, continuously extract target components.
·Key steps/parameters: Use a Soxhlet extraction device and extract continuously with ethanol as the solvent for 2.5 hours.
·Advantages and outputs: High extraction rate. Suitable for laboratory research.
Method selection reference:
·If pursuing speed and greenness, microwave method can be preferred.
·If the equipment requirements are low, consider the acid-base salt precipitation method.
·If the focus is on laboratory research and high extraction rates, ethanol reflux method is very suitable.
💊 Dispersion and formulation technology of berberine
Berberine has poor water solubility and directly affects human absorption. Various dispersion techniques have been developed to improve its solubility and bioavailability.
1. Nanoencapsulation technology
·Core technology: Supercritical solution rapid expansion dispersion method (SEDS).
·Function and effect: The drug is made into a nanoscale inclusion complex, which significantly increases the specific surface area, thereby greatly improving the dissolution rate and solubility. Research has confirmed that its effectiveness is superior to traditional packaging compounds and active ingredients.
2. Solid dispersion technology
·Core technology: Using polymer materials such as PEG6000 as carriers to highly disperse drugs.

·Function and effect: Achieve rapid disintegration and drug dispersion, easy to take. The optimized disintegrant combination for research is: 15% cross-linked polyvinylpyrrolidone (PVPP), 5% microcrystalline cellulose (MCC), and 4% low substituted hydroxypropyl cellulose (L-HPC).

5、 The process of grinding and dispersing machine
