Extracellular vesicles (EVs) are membrane vesicles secreted by various cells, which participate in intercellular communication by transferring substances such as proteins, metabolites, and nucleic acids. A key consideration in the clinical development of EVs is the selection of traditional EV separation methods. Supercentrifugation requires a long time and a lot of effort, even for large-scale research and inexperienced researchers, but they prefer a fast and efficient way to separate EVs and obtain the desired yield and appropriate particle size. Therefore, a method that can quickly separate high-yield EVs will greatly promote such applications.
A common method for separating EVs is to use precipitation agents such as polyethylene glycol (PEG) combined with centrifugation to obtain EVs for subsequent processing. The precipitation method has several advantages; (i) Frequently used, (ii) using conventional laboratory equipment, (iii) suitable for small and large sample sizes, (iv) having high yield. However, it also has limitations: (i) Polymer pollutants, (ii) longer processing times, (iii) complex cleaning steps, (iv) impact downstream analysis and quantification.
The ExoPRISM EV separation kit, as a new precipitation method, overcomes these obstacles by using low molecular weight electrolytes instead of PEG polymers. (i) EV samples separated and enriched by ExoPRISM are free of polymer contaminants. (ii) Due to the rapid reaction of our reagents and samples within 10 minutes, the overall processing time has been greatly reduced. (iii) ExoPRISM reagent can be easily removed from enriched EVs and can be eluted with the buffer solution of your choice. (iv) Therefore, ExoPRISM has no impact on downstream analysis or application.
This kit includes two types of reagents: reagent B and reagent C, which have been optimized for CCS and urine samples, respectively. Reagent B is the EV precipitation solution, and reagent C is the washing solution.
Ingredient List

Reagent B and reagent C should be stored at room temperature. If there is a precipitate in reagent B, please heat it at 37 ℃ in a stirred incubator until the precipitate disappears with the naked eye. After cooling at room temperature, use it.

Use of reagents
Calculate the required reagent volume based on a 5ml sample. Therefore, the amount of reagents needed should be adjusted according to the sample size.
